The Usability of Korean Rabbit Serum as A Complement in the Test of Microcytotoxicity

Abstract
This study-was carried out to determine the usability of Korean rabbit serum as a complement in the test of HLA. Thirty albino male rabbits, weighing average 2.5kg were purchased from the market. They were kept in an outdoor rabbit house and were fed ordinary rabbit diet for 7 days. Each was fasted overnight prior to bleeding of 20m1 from ear vein. The blood samples were clotted separately to avoid hemolysis. The serum is centrifuged at 4¡ÆC for removal of residual RBC and stored at -70¡ÆC. No special treatment took place during the bleeding and handling. 15 rabbits were selected from 30 which had been tested. for antihuman factors in microcytoxicity test, and the result was negative. Average CH50 of15 rabbits in the test of hemolytic complement activity was 0. 29. Among these 15 rabbits serum, 13, sample showed positive at the dilution of 1 : 1 and 1 : 2 through the microcytotoxicitytest of Terasaki with ABC tray. Two rabbit serum showed availability as a complement for DR tray. In accordance with this result, it was determined that Korean rabbit serum could be used for HLA test as a complement followed by the tests for anti-human factor and hemolytic complement activity.
